Description: The CD23 antigen is the low affinity IgE Fc receptor, which is a 45-49 kDa protein with 38 and 28 kDa fragments. It is expressed on most mature, conventional B cells (but not on peritoneal CD5+ B cells) and can also be found on the surface of T cells, macrophages and platelets. It is distinct from the high affinity IgE receptors found on basophils and mast cells which mediate allergic reactions. The low affinity receptors are thought to play a role in isotype specific immunoregulation. The regulation of CD23 surface expression appears to be integral with the complex IgE system which involves interactions of cells, cytokines, antibodies and regulatory factors. CD23 has been described as a “membrane bound cytokine," in that the soluble cleavage products of CD23 are themselves able to act as cytokines in vitro.

Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Description: Murine CD38, a type II transmembrane glycoprotein, is a bifunctional ectoenzyme capable of catabolizing nicotinamide adenine dinucleotide (NAD+) to cyclic ADP-ribose (cADPR), and then hydrolyzing cADPR to adenosine diphosphoribose (ADPR). It is expressed at high levels on the surface of peripheral B-lineage cells and at low density on germinal center B cells from unimmunized mice. It has also been reported to be expressed at moderate levels on NK cells, a proportion of peripheral T cells, and a subpopulation of thymocytes which are mostly TCRαβ+, CD4-, CD8-. Murine CD38 is also expressed by all Mac-1+ macrophages in the peritoneal cavities of unimmunized mice but not by unstimulated bone-marrow-derived macrophages. Monoclonal antibodies to CD38 have been shown to induce B and T cell proliferation, protect B cells from apoptosis, and inhibit B lymphopoiesis.

Description: The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor α chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. By itself, CD25 binds IL-2 only with low affinity. However, CD25 associates with CD122 (IL-2 receptor β chain) and CD132 (common γ chain) to form the high affinity IL-2 receptor. Description: VLA-4 belongs to the β1-integrin family, which functions both as cell-matrix and cell-cell receptors. As a matrix receptor VLA-4 binds to an alternatively spliced domain of fibronectin, while as a cellular receptor it associates with its counter-receptor ligand VCAM-1. In addition to CD44/Pgp-1, VLA-4 has been shown to be important for lymphopoiesis, and is also believed to be important for extravasation of lymphocytes into sites of inflammation as a second signal in addition to LFA/ICAM-1. CD49d is expressed in low amounts on resting lymphocytes and monocytes, is absent or minimally detectable on stromal cells, whereas virtually all lymphoid and myeloid cells in bone marrow and in long-term culture are positive. Antibody PS/2 also reacts with human VLA-4.
