Description: DZIP1 (DAZ interacting protein 1) is also known as DZIP or DZIP2 and is a 867 amino acid protein which is expressed as three isoforms, designated DZIPb, DZIPt1 and DZIPt2. DZIP1 is localized to testis, oocytes, ovary and fetal brain, as well as in embryonic stem cells and germ cells. In testis, DZIP1 is localized to the nucleus and also shows some cytoplasmic distribution in spermatogonia. DZIP1 belongs to the C2H2-type zinc-finger protein family, and, characteristc of the C2H2-type zinc-finger protein family, DZIP1 contains one C2H2-type zinc-finger region through which it is thought to interact with DAZ, an interaction that promotes spermiogenesis. DZIP1 expression is not found in those afflicted with Sertoli cell-only syndrome (characterized by the absence of germ cells in the testis), suggesting that the lack of DZIP1 may be involved in the pathogenesis of Sertoli cell-only syndrome-induced male sterility.

Description: Zic2 is a C2H2 zinc finger transcription factor that influences forebrain development. Zic2 is a transcriptional repressor and may regulate tissue specific expression of dopamine receptor D1. Zic2 transcript is abundant in the dorsal neural tube/spinal cord, and in the hindbrain. A polyhistidine tract polymorphism in this gene may be associated with increased risk of neural tube defects. This gene is closely linked to a gene encoding zinc finger protein of the cerebellum 5, a related family member on chromosome 13.

Description: GIOT-1 is a 563 amino acid protein belonging to the Krüppel C2H2-type zinc-finger protein family. Localized to the nucleus, GIOT-1 is widely expressed in tissues, with highest levels in liver, kidney, small intestine, pancreas and thymus. GIOT-1 contains 12 C2H2-type zinc fincers and one KRAB domain. Because the KRAB domain functions as a transcriptional repressor when attached to the template DNA, GIOT-1 is thought to be involved in transcriptional regulation. The gene encoding GIOT-1 is localized to chromosome 19q13.12 and two isoforms of GIOT-1 exist as a result of alternative splicing events.

Description: The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF215 (ring finger protein 215), is a 377 amino acid multi-pass membrane protein containing one RING-type zinc finger. The gene encoding RNF215 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

Description: Midline-2 is a 715 amino acid protein encoded by the human gene MID2. Midline-2 belongs to the TRIM/RBCC family and contains two B box-type zinc fingers, one B30.2/SPRY domain, one COS domain, one fibronectin type-III domain and one RING-type zinc finger. Midline-2 is a cytoplasmic protein found as a homodimer or heterodimer with Midline-1. It also interacts with IGBP1 (Lymphocyte signaling protein A4). Dimerization is mediated by the tripartite motif (RBCC; RING- and B box-type zinc fingers and coiled coil domains) and microtubule association is dependent on the C-terminal B30.2 domain. Midline-2 is expressed at low levels in fetal kidney and lung, and in adult prostate, ovary and small intestine.

Description: Ubiquitination is an important mechanism through which three classes of enzymes act in concert to target short-lived or abnormal proteins for destruction. The three classes of enzymes involved in ubiquitination are the ubiquitin-activating enzymes (E1s), the ubiquitin-conjugating enzymes (E2s) and the ubiquitin-protein ligases (E3s). ZNRF2 (zinc and ring finger 2), also known as RNF202, is a 242 amino acid peripheral membrane protein that contains one RING-type zinc finger and localizes to the lysosome, as well as the endosome and the cell junction. Expressed at high levels in brain tissue, ZNRF2 is thought to function as an E3 ubiquitin-protein ligase that may be involved in the establishment and maintenance of neuronal transmission and plasticity. Upon DNA damage, ZNRF2 is subject to phosphorylation, probably by ATR or ATM.

Description: ZNF217 (Zinc-finger protein 217) is a Kruppel-like zinc-finger protein which functions as a transcriptional repressor protein and forms a complex with a number of proteins including CoREST, BHC110/LSD1, HDAC2 and CtBP1. ZNF217 has been shown to repress E-cadherin promoter activity. ZNF217 is overexpressed in a number of cancers including breast and ovarian. ZNF217 and its associated proteins have been implicated in a novel pathway that is involved in cancer progression.

Description: Transcription Factor AREB6 / transcription factor 8 /zinc finger homeodomain enhancer-binding protein inhibits interleukin-2 (IL-2) gene expression. May be responsible for transcriptional repression of the IL-2 gene. Enhances or represses the promoter activity of the ATP1A1 gene depending on the quantity of cDNA and on the cell type.ZEB1 encodes a zinc finger transcription factor that represses T-lymphocyte-specific IL2 gene (MIM 147680) expression by binding to a negative regulatory domain 100 nucleotides 5-prime of the IL2 transcription start site (Williams et al., 1991 [PubMed 1840704]).

Description: KLHL28 is a 571 amino acid protein similar to the Drosophila kelch protein. KLHL26 contains six kelch repeats and one BTB (POZ) domain. The BTB (broad complex, tramtrack and bric-a-brac) domain, also known as the POZ (Poxvirus and zinc finger) domain, is an N-terminal homodimerization domain that contains multiple copies of kelch repeats and/or C2H2-type zinc fingers. Proteins that contain BTB domains are thought to be involved in transcriptional regulation via control of chromatin structure and function. KLHL28 is expressed as two isoforms produced by alternative splicing.
