Catalog Number: (10414-104)
Supplier: Bioss
Description: Two HSFs have been identified in human cells, HSF 1 and HSF 2, which bind to the same HSEs and have 38% sequence identity. These factors are activated by distinct stimuli, HSF 1 is responsive to classical stress signals such as heat, heavy metals and oxidative reagents, whereas HSF 2 is activated during hemin-mediated differentiation of human erythroleukemia cells.HSF 1 exists constitutively in the cytoplasm and the nucleus of unstressed cells as a monomer which lacks DNA binding activity. Through an unknown signal generated during stress, HSF 1 becomes activated to a nuclear localized, trimeric state which binds to DNA. The phosphorylation of HSF 1 is necessary for maximal transcription of heat shock genes.

Catalog Number: (10492-810)
Supplier: Bioss
Description: JWA is a four-transmembrane environmental responsive protein which binds to the CC chemokine recepor 5 (CCR5), a major co-receptor for human immunodeficiency virus (HIV). JWA is involved in environmental stress-responsive pathways in K562 cells, an erythroleukemia cell line derived from patients with chronic myeloid leukemia. Environmental stressors to K562 cells such as heat shock, a higher temperature than the ideal body temperature of the organism from which the cell line was derived, and oxidative stress, the production of oxygen-centered free radicals, regulate and increase the expres-sion of JWA. This response to environmental stressors suggests similiarity of JWA to heat shock protein 70 (HSP70), which is upregulated by heat stress and toxic chemicals.

Catalog Number: (75928-584)
Supplier: Rockland Immunochemical
Description: HSF1, or heat shock factor 1, belongs to a family of Heat Shock transcription factors that activate the transcription of genes encoding products required for protein folding, processing, targeting, degradation, and function (2). The up-regulation of HSP (heat shock proteins) expression by stressors is achieved at the level of transcription through a heat shock element (HSE) and a transcription factor (HSF) (3, 4, 5). Most HSFs have highly conserved amino acid sequences. On all HSFs there is a DNA binding domain at the N-terminus. Hydrophobic repeats located adjacent to this binding domain are essential for the formation of active trimers. Towards the C-terminal region another short hydrophobic repeat exists, and is thought to be necessary for suppression of trimerization (6). There are two main heat shock factors, 1 and 2. Mouse HSF1 exists as two isoforms, however in higher eukaryotes HSF1 is found in a diffuse cytoplasmic and nuclear distribution in un-stressed cells. Once exposed to a multitude of stressors, it localizes to discrete nuclear granules within seconds. As it recovers from stress, HSF1 dissipates from these granules to a diffuse nuceloplasmic distribution. HSF2 on the other hand is similar to mouse HSF1, as it exists as two isoforms, the alpha form being more transcriptionally active than the smaller beta form (7, 8). Various experiments have suggested that HFS2 may have roles in differentiation and development (9, 10, 11). Anti-HSF1 Antibody is ideal for research in Genetics, Transcription, Cell Signaling and pathways including ERK and MAPK.
