Description: SAE2 Antibody: Small ubiquitin-like modifiers (SUMOs) are a family of small, related proteins (SUMO-1/2/3/4) that can be enzymatically attached to a target protein by a post-translational modification process termed sumoylation, a major regulator of protein function in cellular processes such as nuclear transport, transcriptional regulation, apoptosis and protein stability. This sumoylation is effected by the heterodimeric enzyme SAE1/SAE2 and the SUMO-1-conjugating enzyme Ubch9. The sumoylation pathway mediated by SAE1/SAE2 is distinct from other ubiquitin-like protein (Ubl) pathways.

Description: SUMO3 Antibody: Small ubiquitin-like modifiers (SUMOs) are a family of small, related proteins (Sumo-1/2/3/4) that can be enzymatically attached to a target protein by a post-translational modification process termed sumoylation, which is a major regulator of protein function in cellular processes such as nuclear transport, transcriptional regulation, apoptosis and protein stability. All SUMO proteins localize to the nucleus and are covalently conjugated, affecting protein structure, function and interactions. SUMO2 and 3 are 96% identical and are more mobile within nucleus relative to SUMO1. Specific functional differences between SUMO1 and SUMO2 and 3 remain to be identified.

Description: CDK5 (Cyclin Dependent Kinase 5) is serine/threonine kinase involved in synaptic regulation and neuronal development; phosphorylates synaptic protein Pctaire1; regulates acetylcholine receptor expression. CDK5 is a member of the cyclindependent kinase family of serine/threonine kinases. It is present in numerous mammalian tissues including kidney, testes, and ovary. Its activity is detected almost exclusively in brain extracts. Neuronal and muscle cells contain the highest amount of this protein. Similar to other Cdks, monomeric Cdk5 displays no enzymatic activity, but Cdk5 is not activated by cyclins. Instead, the p35 protein, which is expressed solely in the brain, activates Cdk5. Cdk5 interacts with D1 and D3 type G1 cyclins and can phosphorylate histone H1, TAU, MAP2 and NF-H and NF-M. Cdk5 activity is involved in terminal differentiation of neurons and muscle cells.

Description: SH3RF2 Antibody: Although the SH3 domain containing ring finger 2 protein (SH3RF2) has been identified as a marker of the nucleus accumbens in the human brain, little is known about this protein. Recent yeast two-hybrid experiments have shown that SH3RF2 associates with protein phosphatase 1 (PP1) and is expressed primarily in heart and testis tissue. Recombinant SH3RF2 enhanced PP1 enzymatic activity and antagonized the ability of phopho-inhibitor-1 or -2 to inhibit PP1, suggesting that SH3RF2 may play a role working with PP1 in cardiac functions.

Description: Cyclic AMP-dependent phosphodiesterase type D (PDE4D) family is comprise of 5 variants (PDE4D1, D2, D3, D4 and D5). One or more PDE4D subtype-variants are ubiquitously present in all mammalian cells. In CNS all five PDE4D subtype-variants are expressed in varying ratios and their activity is regulated in tandem with GPCRs stimulation. Peripheral tissues also exhibit differential expression of PDE4D variants. PDE4D1/D2 mRNA levels rise in response to an increase in cAMP. Short-term regulation of PDE4D variants involved PKA, MAP kinases and Erk2 phosphorylation that results in rapid change in their enzymatic activities. Other regulatory mechanism involved protein-protein interactions with cytoskeletal scaffolding proteins.

Description: SIRT7 is a human member of a family of proteins called Sirtuins (Sir2-like proteins) and are present in prokaryotes and eukaryotes. All Sir2-like proteins have a sirtuin core domain, which contains a series of sequence motifs conserved in organisms ranging from bacteria to humans. Bacterial, yeast and mammalian sirtuins are able to metabolize NAD and possibly at as mono-ADP-ribosyltransferases. The enzymatic function of sirtuins is not yet completely understood but recent reports of histone-activated Sir2-mediated NAD metabolism and NAD-activated Sir2-mediated histone deacetylation suggest a possible coupled reciprocal activation mechanism involving interactions of Sir2 with NAD and the N epsilon-acetyl-lysine groups of acetylated histones.

Description: Apoptosis is regulated by death domain (DD) and/or caspase recruitment domain (CARD) containing molecules and a caspase family of proteases. CARD containing cell death regulators include RAIDD, RICKBCL10, Apaf-1, caspase-9, and caspase-2. A novel CARD domain containing protein was recently identified and designated ARC for apoptosis repressor with CARD (1). ARC interacts withcaspase-2 and ~8 and inhibits enzymatic activity ofcaspase-8. ARC suppresses apoptosis induced by cell death adapters FADD and TRADD and by cell death receptors Fas, TNFR-1, and DR3. The messenger RNA of ARC is primarily expressed in skeletal muscle and cardiac tissue (1).

Description: The class III type phosphoinositide 3-kinase (PI3KC3) / Vps34 regulates vacuolar trafficking as well as autophagy. UVRAG (UV radiation resistance-associated gene) is associated with the Beclin-1/PI3KC3 complex and promotes PI3KC3 enzymatic activity and autophagy, while suppressing proliferation. UVRAG is highly expressed in brain, lung, kidney and liver and contains one C2 domain, which is involved in calcium-dependent phospholipid binding. Beclin-1 binding to UVRAG promotes both autophagosome maturation and endocytic trafficking. UVRAG is also a potential tumor suppressor protein with frameshift mutations observed in colon and gastric carcinomas.

Description: FUNCTION: Required for T-cell activation through the antigen receptor. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one.
DISEASE: Defects in PTPRC are a cause of autosomal recessive severe combined immunodeficiency T cell-negative/B cell-positive/NK cell-positive (T(-)/B(+)/NK(+) SCID) [MIM:608971]. SCID refers to a genetically and clinically group of rare congenital disorders characterized by impairment of both humoral and cell-mediated immunity, leukopenia, and low or absent antibody levels. Patients with SCID present in infancy with recurrent, persistent infections by opportunistic organisms, including Candida albicans, Pneumocystis carinii, and cytomegalovirus, among many others. The common characteristic of all types of SCID is absence of T cell-mediated cellular immunity due to a defect in T-cell development.
