Description: In muscle tissue, collagen serves as a major component of the endomysium. Collagen constitutes one to two percent of muscle tissue, and accounts for 6% of the weight of strong, tendinous muscles. A collagen may be defined as a protein containing sizable domain(s) of triple-helical conformation. Type IV collagen is a major macromolecular constituent of basement membranes and can be readily isolated from basement-membrane-rich tissues or highly vascularized tissues such as the placental villi. This collagen appears to be largely restricted to structures identifiable as basement membranes. In contrast, type VI collagen appears to be prevalent in several tissues even though it has been isolated largely from placental villi preparations. The extent to which type VII and VIII collagens are distributed is not known.

Description: BRCC45 was initially suggested to be a housekeeping protein that is highly expressed in brain and reproductive organs. Later experiments indicated BRCC45 forms a complex with the breast and ovarian predisposition proteins BRCA1 and BRCA2 as well as RAD51 and BRCC36. This complex has a ubiquitin E3 ligase activity and is thought to enhance cellular survival following DNA damage. BRCC45 has also been suggested to function as a death receptor-associated anti-apoptotic protein by inhibiting the BID-induced activation of the mitochondrial apoptotic pathway. Higher levels of BRCC45 were detected in the majority of hepatocellular carcinomas, suggesting that BRCC45 may promote tumorigenesis when overexpressed. Description: DARC, also known as the Duffy antigen/chemokine receptor, is a seven-transmembrane protein homologous to the classical chemokine G-protein coupled receptors (GPCRs) with the exception of the motif required for G protein coupling. DARC can bind with high affinity several chemokines without transducing any signal, suggesting it may modulate the signals normally induced by these chemokines. Recently, DARC was found to interact with KAI1, a four transmembrane protein recently identified as a tumor metastasis suppressor protein. It is thought that tumor cells dislodged from the primary tumor and expressing KAI1 interact with DARC proteins expressed on vascular cells, transmitting a senescent signal to the tumor cells, while tumor cells that have lost KAI1 expression can proliferate and potentially give rise to metastases. Description: STAT2 is a member of the STAT family of transcription factors. Unlike other STATs, STAT2 is unique as it can only be activated by interferons (IFNs). STAT2 is a critical component in mediating many IFN-stimulated biological activities including antiproliferation and antiviral responses. Upon IFN treatment, STAT1 and STAT2 become tyrosine phosphorylated, assemble as heterodimers that bind IRF9 to form the ISGF3 complex. This complex translocates to the nucleus, binds to promoters of IFN-stimulated genes and mediates gene transcription. Consequently, mutations in STAT2 or loss of STAT2 expression leads to impairment in IFN signal transduction and gene activation. IFN-alpha is an approved drug for the treatment of several forms of cancer. Yet only a subset of patients who receive IFN-alpha therapy benefit from the treatment. Given that STAT2 is activated by IFNs, it is important to define if the reduced or lack of antitumor effects seen in cancer patients on IFN therapy is due to in defects in STAT2 function.
