Description: Disruptions of protein folding and maturation in the endoplasmic reticulum (ER) result in the activation of the unfolded protein response (UPR), an integrated cellular signaling pathway that transmits information from the ER lumen to the cytoplasm and nucleus. Activating transcription factor 6 (ATF6) as well as the ER-transmembrane protein kinases IRE1p and PERK are the major transducers of the UPR. ATF6 is an ER transmembrane protein that is normally bound to the ER chaperone Grp78, but upon ER stress is released from Grp78 and proteolytically cleaved to yield a cytosolic fragment which then migrates to the nucleus. Nuclear ATF6, together with the transcription factor XBP-1, activates transcription of UPR-responsive genes.

Description: Influenza A virus is a major public health threat, killing more than 30, 000 people per year in the USA. Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals; however, it is in birds that all subtypes, including the so-called "avian flu" or H5N1, can be found. These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. One of the less studied proteins encoded by, but not incorporated in, the influenza virus is the nonstructural protein (NS) 1. NS1 counters cellular antiviral activities and acts as a virulence factor. It can bind to double-stranded RNA and sequester it from 2'-5'OAS, preventing the activation of the RNAse L, which normally acts to degrade RNA and prevent virus replication. NS1 also binds to and inhibits the anti-viral protein kinase PKR.

Description: CKIP1 was identified through a yeast-two hybrid screening as a protein that would interact with only one of the two catalytic subunits of the casein kinase 2 complex. CKIP1 is a pleckstrin homology domain-containing protein localized within the nucleus and at the plasma membrane that interacts with CK2alpha but not CK2alpha', and is thought to play a role in targeting CK2alpha to a particular cellular location. CKIP1 has been implicated in muscle differentiation and the regulation of cell morphology and actin cytoskeleton. CKIP1 can also interact with other proteins such as ATM, an upstream kinase of p53, and recruit the nuclear ATM to the plasma membrane, suggesting CKIP1 may regulate ATM function through re-localizing ATM.
