Description: The JES6-5H4 antibody binds to mouse Interleukin-2 (IL-2), a 17 kDa cytokine that is secreted by activated T cells. This cytokine acts as an autocrine stimulatory factor for T cell proliferation, differentiation and survival, as well as being an effective activator of B cells and inducer of NK cell cytotoxic functions. IL-2 plays a key modulatory role in the differentiation of T cells toward either Th17 or T regulatory (Treg) cell types and is important for Treg cell function. The IL-2 receptor (IL-2R) is a complex consisting of an IL-2R alpha chain (CD25), along with the IL-2R beta chain (CD122) and the common gamma chain (CD132). IL-2 may also bind with low affinity to the T cell surface protein CD25 alone, although this interaction does not induce cell signaling.

Description: Glycogen Synthase Kinase 3β (GSK-3β) is a unique serine/threonine kinase that is inactivated by phosphorylation. In response to insulin binding, PKB/AKT phosphorylates GSK-3β on serine 9, which prevents GSK-3β from phosphorylating glycogen synthase. Unphosphorylated glycogen synthase is active and able to synthesize glycogen. GSK-3β is also unique in that it requires a substrate that has been phosphorylated by a distinct kinase before it can phosphorylate the substrate. This phosphate priming mechanism explains why phosphorylation of serine 9 inactivates GSK-3β. The phosphorylated serine binds to the GSK-3β priming phosphate position and prevents binding of alternative substrates. In addition to insulin signaling, GSK-3β participates in the Wnt signaling pathway, where it forms a complex with axin, beta-catenin and adenomatous polyposis coli (APC) protein. In the presence of Wnts, GSK-3β is unable to phosphorylate beta-catenin, which leads to stabilization of beta-catenin. The Wnt pathway inactivates GSK-3β via the proteins, Dishevelled and FRAT, which disrupt the interaction of GSK-3β with axin, beta-catenin, and APC. Clinically, there is considerable interest in GSK-3β inhibitors because they may mimic the effect of insulin or reduce the hyperphosphorylation of Tau that is observed in Alzheimer's Disease.

Description: Mitochondria antibody detects human Mitochondria. Mitochonria are most commonly known as the power plants of the cell as they produce ATP, but they are also involved in many other important cellular processes such as cell signaling, growth and differentiation. In addition, mitochondria have been shown to play a role in apoptosis. This antibody is an excellent marker for human cells in xenographic model research.

Description: Bora (Protein aurora borealis) is a key activator of Aurora Related Protein Kinase A (ARK-1), which is a centrosome-associated serine/threonine kinase that regulates centrosome maturation, bipolar spindle assembly and chromosome segregation during mitosis. Bora is localized to the nucleus until mitosis is initiated, then translocates to the cytoplasm in a Cdc2 dependent manner. Activation of Cdc2 initiates the release of Bora into the cytoplasm where it can bind and activate ARK-1. PLK1 (polo-like kinase-1) interacts with Bora to control the accessibility of its activation loop for phosphorylation and activation by ARK-1. Bora and ARK-1 cooperatively activate PLK1 and control mitotic entry. Bora mutants result in multipolar spindles in mitosis identical to those observed when ARK-1 function is blocked. Thus, the ARK1-Bora-PLK1 regulatory circuit in mammalian cells elucidates a key mechanism in cell cycle regulation. At least three isoforms of Bora are known to exist.

Description: The BM8.1 antibody is specific for mouse F4/80 antigen, a 125 kDa transmembrane protein widely expressed by members of the mononuclear phagocyte system and considered to be a key marker for mature macrophage cells. F4/80 is differentially expressed during myeloid cell development, and may be regulated by certain cytokines within the tissue microenvironment. Other cell types shown to express this antigen include Langerhans cells, Kupffer cells and dendritic cell subsets. BM8.1 is widely used together with antibodies to CD115 (c-fms), CD11b and CD11c to identify myeloid / macrophage cells by flow cytometry.

Description: The multifunctional, multi-compartmental protein Calreticulin (Crt) functions as a soluble molecular chaperone of new or misfolded proteins as well as a Ca2+-binding protein. Most abundant in the ER lumen, Crt expression also occurs in other membrane-bound organelles, the cell surface, and extracellularly. Also known as CRP-55, calregulin and HACBP (high affinity calcium-binding protein), Crt contains the ER-retrieval sequence, KDEL, and is the soluble paralog of the ER membrane protein Calnexin (Cnx). Crts three domains include a 180 residue N-terminal domain, a proline-rich P-domain (residues 189-288) that binds Ca2+ with high affinity and shares homology with Cnx and calmegin, and a 110 residue C-terminal domain that binds Ca2+ with low affinity but high capacity. The P-domain may interact with the co-chaperone ERp57 (Grp58), a thiol reductase. The NMR structure of the P-domain consists of an extended hairpin that appears to form a curved protrusion from the Crt core domain. Both Crt and its membrane bound homolog CNX interact with proteins and glycoproteins possessing monoglucosylated N-glycans. The Crt/Cnx cycle promotes correct folding, inhibits aggregation of folding intermediates, blocks premature oligomerization, regulates ER degradation, and prevents incompletely folded glycoproteins from exiting to the Golgi complex. Crt also appears to function as an auto-antigen in systemic lupus erythematosus, rheumatoid arthritis, celiac disease, complete congenital heart block, and halothane hepatitis. A diversity of additional functions attributed to Crt includes adhesion, blood function, and cardiac and neuronal development gene expression.

Description: Plxdc2, also known as Tumor endothelial marker 7-related (TEM7R) encodes a protein with 57% amino acid identity to TEM7, the most abundant tumor endothelial marker. Plxdc2 is strongly expressed in the endothelial cells of the tumor stroma, but not in the endothelial cells of normal colonic tissue. Plxdc2 is also expressed at high levels in vessels of some normal tissues, with highest expression in muscle and lung. Plxdc2 and TEM7 may be important for tumor angiogenesis in humans. Cortactin was identified as a protein capable of binding to the extracellular region of both TEM7 and Plxdc2, and may provide new opportunities for the delivery of therapeutic and imaging agents to the vessels of solid tumors.

Description: Nudel was initially discovered as a protein homologous to Aspergillus NUDE and that associated with Lis1 and polyprotein complex cytoplasmic dynein, both of which have been linked to neuronal development and migration. It was later shown to be a substrate of cdk5, a kinase known to be critical during neuronal migration; phosphorylation of Nudel by cdk5 affects its localization in neurons and affects neuritic morphology. It is thought that together with Lis1, Nudel regulates cytoplasmic dynein, a large polyprotein complex, in neuronal migration and mitosis through direct interactions. Similar interactions are thought to also play a role in membrane traffic in other cells as disruption of Nudel expression through RNA interference perturbed normal endomembrane flux and resulted in the fragmentation of the Golgi apparatus.

Description: The ectoderm-neural cortex-1 (ENC-1) protein is an early and highly specific marker of neural induction in vertebrates. It is a kelch family related protein that functions as an actin-binding protein and has been suggested to be involved in the organization of the actin cytoskeleton during neural fate specification and development of the nervous system. ENC-1 has also been shown to be required for adipocyte differentiation when cytoskeletal reorganization and cell shape change from fibroblastic preadipocytes to spherical adipocytes occur.

Description: Rockland produces a wide range of human GST antibodies in our laboratories. Select appropriate GST antibodies for your research by isotype, epitope, applications and species reactivity. There are 22 members of the human GST family of proteins. GST is responsible for the conjugation of reduced glutathione to a wide number of exogenous and endogenous hydrophobic electrophiles. The amino acid sequence GST is highly conserved in most organisms including mammals. GSTs proteins are typically homodimeric, with both heterologous GST dimers have been observed. GST monomers have an average molecular weight of approximately 25-28 kDa in size. Note a different form of non-human GST (Glutathione-S-Transferase) is used as a protein expression tag commonly in molecular biology applications. All anti-GST antibodies my not react with recombinant GST-fusion proteins.
