Description: CD138, also known as Syndecan-1, is a member of the transmembrane heparan sulfate proteoglycan family, acts as an extracellular matrix receptor and is involved in many cellular functions, including cell-cell adhesion and cell-matrix adhesion. CD138 expression is found in both hematopoietic and non-hematopoietic cells. In the hematopoietic system, CD138 labels plasma cells. It is an excellent marker for plasmacytic differentiation within the spectrum of hematologic malignancy. Among non-hematolymphoid cells, CD138 reactivity is observed in many types of epithelial cells and stoma cells in both normal and tumor tissues.

Description: The mitochondrial succinate dehydrogenase complex subunit D (SDHD) is one of four proteins that make up the tricarboxylic cycle enzyme succinate dehydrogenase (SCH). Studies have shown that mutations in SDHD often leads to hereditary paragangliomas, usually benign tumors of the autonomic nervous system, suggesting that SDHD also plays a role as a tumor-suppressor gene. In one family with a nonsense mutation (R22X) in the SDHD gene, a loss of heterozygosity was found in the paragangliomas, and within these tumors the enzymatic activity of Complex II in the mitochondrial respiratory chain was completely abolished. Furthermore, high levels of angiogenic factors EPAS1 and VEGF was observed, which may stimulate tumor growth.

Description: The SK3 antibody reacts with human CD4, a 59 kDa protein which acts as a co-receptor for the T cell receptor (TCR) in its interaction with MHC Class II molecules on antigen-presenting cells. The extracellular domain of CD4 binds to the beta-2 domain of MHC Class II, while its cytoplasmic tail provides a binding site for the tyrosine kinase lck, facilitating the signaling cascade that initiates T cell activation. CD4, and co-receptors CCR5 and CXCR4, may also be utilized by HIV-1 to enter T cells. Human CD4 is typically expressed on thymocytes, some mature T cell populations such as Th17 and T regulatory (Treg) cells, as well as on dendritic cells. The SK3 antibody is widely used as a phenotypic marker for human CD4 expression, and has been reported to be cross-reactive with Rhesus and Cynomolgus CD4. This antibody does not block binding of alternative clone RPA-T4, suggesting that they recognize different epitopes.

Description: The RA3-6B2 antibody reacts with the human and mouse CD45 isoform known as CD45R, or B220, a protein tyrosine phosphatase of 220 kDa. CD45 is one of the most abundant hematopoietic markers, and is expressed on all leukocytes (the Leukocyte Common Antigen, LCA). Various isoforms are generated and expressed in cell-specific patterns, all critical for leukocyte function. In mouse, the CD45R/B220 isoform is predominantly found on B cells, at varying levels on all stages from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. It is of note that B220 is not similarly expressed on human B cells, where it appears to be differentiation-specific and therefore expressed on only some B cell subsets. Description: Heme Oxygenase-1 (HO-1) also known as Hsp32, is the inducible isoform of heme oxygenase that catalyzes the NADPH, oxygen, and cytochrome P450 reductase dependent oxidation of heme to carbon monoxide, ferrous iron and biliverdin which is rapidly reduced to bilirubin. These products of the HO reaction have important physiological effects: carbon monoxide is a potent vasodilator and has been implicated to be a physiological regulator of cGMP and vascular tone; biliverdin and its product bilirubin are potent antioxidants; "free" iron increases oxidative stress and regulates the expression of many mRNAs (e.g., DCT-1, ferritin and transferring receptor) by affecting the conformation of iron regulatory protein (IRP)-1 and its binding to iron regulatory elements (IREs) in the 5'- or 3'- UTRs of the mRNAs. To date, three identified heme oxygenase isoforms are part of the HO system that catalyze heme into biliverdin and carbon monoxide.
